For 4u specifically, it's always recommended to complete the story mode first, because it unlocks a lot of useful stuff that you won't get access to otherwise (like a big portion of the canteen, the trader and the farm (wycoon), decorations, wystones etc) I remember in mh4u you could only unlock wystones and having double wystones by playing the single player campaign and it was such a great incentive to play solo as well as show that you had the skill to beat the monsters without help. Unlock first Wystone: Do the quest "Ladykiller" in High Rank Caravan Unlock second Wystone slot: Do quests "Advanced: Tiger-sharq" G1 and "Advanced: Escaped Convicts" G2 then a Wyporium Villager Request will open up, complete it to unlock 2nd slot.
